Induction and identification of tetraploids of pear plants (Pyrus bretschneideri and Pyrus betulaefolia)

چکیده

Polyploid plants usually exhibit broader leaves, thicker stems, bigger flowers and fruits, dwarfing stature, as well improved biotic abiotic resistance. Therefore, increasing the polyploidy is one of most important strategies used in plant breeding. Here, we reported successful induction tetraploids two pear varieties, ‘duli’ (Pyrus betulaefolia), a wildtype for rootstocks grafting, ‘Xinli No.7′ bretschneideri), popular cultivated variety China. This was achieved by treating their seeds, shoot tips tissue cultured or field grown seedlings with colchicine pendimethalin using three different methods: impregnation, mixed culture smearing. The best condition seeds impregnating them 0.4% 24 h where mutation rate 2.0% no chimera found. For 0.2% 6.67% (48 h) 13.0% (24 were mutated. In addition, found that method mixing 1.0% while it 0.5% colchicine. smear method, ‘duli’, h. Subsequently, DNA content these putative polyploid estimated flow cytometry, confirming ploidy nature. Also, compared morphological differences between tetraploid diploid seedlings. Obviously, appeared to be dwarfed shorter internode length reduced number stems.

Sequence Characterization and Spatiotemporal Expression Patterns of PbS26-RNase Gene in Chinese White Pear (Pyrus bretschneideri)

Many flowering plants exhibit an important intraspecific reproductive barrier phenomenon, that is, self-incompatibility (SI), in which S-RNase genes play a significant role. To clarify the specific function of S-RNase genes in Chinese pears, the full length cDNA of PbS 26 -RNase was isolated by rapid amplification of cDNA ends (RACE) technology from Chinese white pear (Pyrus bretschneideri) cul...

Genome-wide identification of the MADS-box transcription factor family in pear (Pyrus bretschneideri) reveals evolution and functional divergence

MADS-box transcription factors play significant roles in plant developmental processes such as floral organ conformation, flowering time, and fruit development. Pear (Pyrus), as the third-most crucial temperate fruit crop, has been fully sequenced. However, there is limited information about the MADS family and its functional divergence in pear. In this study, a total of 95 MADS-box genes were ...

Identification of Pyrus Single Nucleotide Polymorphisms (SNPs) and Evaluation for Genetic Mapping in European Pear and Interspecific Pyrus Hybrids

We have used new generation sequencing (NGS) technologies to identify single nucleotide polymorphism (SNP) markers from three European pear (Pyrus communis L.) cultivars and subsequently developed a subset of 1096 pear SNPs into high throughput markers by combining them with the set of 7692 apple SNPs on the IRSC apple Infinium® II 8K array. We then evaluated this apple and pear Infinium® II 9K...

MYB Transcription Factors in Chinese Pear (Pyrus bretschneideri Rehd.): Genome-Wide Identification, Classification, and Expression Profiling during Fruit Development

The MYB family is one of the largest families of transcription factors in plants. Although, some MYBs were reported to play roles in secondary metabolism, no comprehensive study of the MYB family in Chinese pear (Pyrus bretschneideri Rehd.) has been reported. In the present study, we performed genome-wide analysis of MYB genes in Chinese pear, designated as PbMYBs, including analyses of their p...

Structural, Evolutionary, and Functional Analysis of the Class III Peroxidase Gene Family in Chinese Pear (Pyrus bretschneideri)

Peroxidases (PRXs) are widely existed in various organisms and could be divided into different types according to their structures and functions. Specifically, the Class III Peroxidase, a plant-specific multi-gene family, involves in many physiological processes, such as the metabolism of auxin, the extension and thickening of cell wall, as well as the formation of lignin. By searching the pear...
